پایش عامل بیماری سوختگی باکتریایی پنبه و بررسی باکتریهای گرم منفی رورست در بذر پنبه
فیلوسفر مکانی برای زندگی ریزموجودات است. باکتریها از جمله ساکنین فراوان فیلوسفر هستند که تعداد آنها تحت تاثیر گونه گیاه و نوع برگ قرار میگیرد. باکتریهای متعددی بصورت رورست در سطح اندامهای هوایی و بذور گیاه وجود داشته و در شرایط مساعد باعث بروز بیماری میشوند. در طی سالهای اخیر با بروز همهگیری بیماری بلایت باکتریائی پنبه در کشور و بخصوص استان گلستان در سطح وسیع نظریاتی مبنی بر وجود رورستی باکتری در سطح بذور پنبه مطرح گردید. به منظور شناسائی باکتریهای گرم منفی موجود در سطح بذور پنبه، ازبذور توصیه شده ارقام تجاری ساحل، ورامین، خرداد، بختگان و سپید بصورت تصادفی نمونه برداری شد. با قرار دادن بذور در داخل آب مقطر سترون و افزودن مقداری توئین 20 به عنوان ماده شوینده و قرار دادن محیط فوق در دمای اتاق به مدت یک ساعت به شستشوی بهتر باکتریهای موجود در سطح بذور کمک شد. سوسپانسیون فوق در روی محیط کشت مناسب مخطط گردید. در ادامه باکتریهای رشد کرده جداسازی، خالصسازی و بر اساس منابع معتبر باکتریولوژی شناسائی شدند. بر اساس نتایج حاصل گونههای Psuodomonas fluorescens، P. syringae، Pantoea annanas و Pectobacterium sp. شناسایی شدند. هیچ موردی از وجود باکتری Xanthomonas citri subsp.malvacearum به عنوان عامل بیماری بلایت باکتریایی بهصورت رورست در بذور پنبه به اثبات نرسید.

مدلسازی ریاضی ویژگی های گرمایی دانه پنبه کرک دار و بدون کرک در سه رقم جدید پنبه کاشمر، لطیف و خورشید در دماهای مختلف
تعیین برخی خواص مختلف گرمایی دانه پنبه مانند ضریب هدایت گرمایی، گرمای ویژه و ضریب پخش گرمایی در فرآیندهای مختلف گرمایی، انبارداری و طراحی خشککنها کاربرد دارد. سه ویژگی مهم گرمایی شامل ضریب هدایت گرمایی، گرمای ویژه و ضریب انتشار گرمایی سه رقم دانه پنبه به نامهای کاشمر، لطیف و خورشید در دو حالت کرکدار و بدونکرک در سه دمای 20، 30 و ˚c40 اندازهگیری گردید. در این پژوهش ضریب هدایت حرارتی دانه پنبه از روش منبع گرمای خطی، گرمای ویژه از روش مخلوط و ضریب انتشار گرمایی با استفاده از نتایج حاصل از آزمایشهای مربوط به ضریب هدایت گرمایی، گرمای ویژه و چگالی توده دانه پنبه محاسبه گردید. نتایج نشان داد که تغییرات دما بر ضریب هدایت گرمایی سه رقم دانه پنبه معنیدار نبود. تغییرات رقم، دما و پوشش در سطح احتمال 1 درصد بر گرمای ویژه موثر بود و همچنین اثر متقابل رقم در پوشش و دما در پوشش بر گرمای ویژه دانه پنبه در سطح احتمال 1 درصد معنیدار بود. بیشترین و کمترین مقدار گرمای ویژه دانه پنبه کرکدار در دمای 40 و ˚c20 بترتیب، 83/1 و (kJ/kg˚c) 18/1 و در دانه پنبه بدون کرک 45/1 و (kJ/kg˚c) 86/0 بود. نتایج آزمایشهای ضریب انتشار گرمایی نشان داد که تغییرات رقم، دما و پوشش در سطح احتمال 1 درصد بر ضریب انتشار گرمایی موثر است و همچنین اثر متقابل رقم در پوشش و دما در پوشش بر ضریب انتشار گرمایی دانه پنبه در سطح احتمال 1 درصد معنیدار بود. بیشترین و کمترین مقدار ضریب انتشار گرمایی دانه پنبه کرکدار در دمای 20 و ˚c40 بترتیب، 7-10×48/3 و (m2/s) 7-10×26/2 و در دانه پنبه بدون کرک 7-10×57/2 و (m2/s) 7-10×53/1 بود.

بررسی ریزریختشناسی و تشریحی ساختار بذر در دو رقم پنبه و اهمیت کاربرد آنها
مطالعه خصوصیات ریزریختشناسی و تشریحی بذرها طیف گستردهای از ویژگیهایی را ارائه میدهد که نقش مهمی در بررسی کیفیت بذر و شناسایی ریزریختشناسی ارقام مختلف را فراهم میکنند. سلامت بخشهای تشکیل دهندهی بذر، برای جوانه زدن و تولید محصول مطلوب، همواره لازم و ضروری است. هدف از این مطالعه مقایسه ساختار تشریحی رویان و بذر دو رقم پنبه (لطیف و گلستان) و اهمیت کاربرد این ویژگیها در زمان کاشت و افزایش عملکرد پنبه بود. در این مطالعه بذرهای هر دو رقم از نظر ویژگیهای مورفومتری بررسی شدند. ساختار تشریحی درونی و ساختار بذرها بعد از مراحل آماده سازی و برشگیری با دستگاه میکروتوم به روشهای مختلف رنگآمیزی، با میکروسکوپ نوری و فلورسنت مطالعه شدند. نتایج این بررسی نشان داد که بذر رقم گلستان سطح، قطر، نسبت اقطار، فاکتور شکل، فاکتور گردی و وزن هزار دانه بیشتری داشت. در رقم گلستان مریستمها کاملا شکل گرفتهاند و پرومریستمها در منطقه هیپوکوتیل نسبت به رقم لطیف توسعه یافتهترند. در هر دو رقم، سلولهای کلرانشیم اسفنجی و نردبانی مزوفیل متمایز شدهاند اما این سلولها در رقم گلستان نمو بیشتری داشتند. در رنگآمیزی تولوئیدن برای شناسایی ترکیبات فنلی، ساختار حفره ترشحی در بذرهای گلستان نسبت به رقم لطیف رنگپذیری بالایی داشتند. میکروسکوپ فلورسنت، حضور فراوان ترکیبات فلوروسنت را در ساختار بذر نشان داد. در نهایت میتوان بیان داشت که رقم گلستان به سبب بلوغ زودرس رویان و درصد جوانه زنی بیشتر، در شرایط یکسان میتواند درصد سبز بیشتری نسبت به رقم لطیف داشته باشد.

تنوع ژنوتیپی و اجزای همبسته با عملکرد، کیفیت الیاف و هتروزیس در برخی هیبریدهای پنبه
بهبود عملکرد یکی از اهداف مهم بهنژادی پنبه محسوب میشود و بهرهگیری از هتروزیس بهعنوان یکی از ابزارهای قدرتمند جهت ارتقای عملکرد و کیفیت الیاف پنبه همیشه مورد توجه بهنژادگران پنبه بوده است. به منظور بررسی خصوصیات والدینی و میزان هتروزیس مفید قابل دستیابی در هیبریدهای پنبه، تلاقیهای متعددی در سالهای 1389 و 1391 انجام و بر اساس ارزیابیهای مقدماتی تعداد ده هیبرید انتخاب و در سال 1393 در کنار ده ژنوتیپ والدینی و تجاری در قالب طرح بلوکهای کامل تصادفی در سه تکرار ارزیابی شدند و دادههای آزمایشی با استفاده از روشهای آماری و بیومتریک مورد تجزیه و تحلیل قرار گرفتند. نتایج تجزیه واریانس صفات دلالت بر تنوع ژنوتیپی و اختلاف معنیدار بین هیبریدها و والدین مورد مطالعه برای اکثر صفات کمی و کیفی داشت. در بین هیبریدهای مورد بررسی، Mehr× BeliIzovar و T2×Bakhtegan بهترتیب با عملکرد 4993 و 4876 کیلوگرم در هکتار در رتبههای اول و دوم قرار گرفتند و میزان هتروزیس عملکرد آنها نسبت به والد برتر مثبت و به ترتیب 7/24 و 8/22 درصد بود. بر اساس عملکرد و شاخصهای کیفیت الیاف، هیبریدهای Sahel× Khordad و Sahel× B557 در رتبههای بالاتر قرار گرفتند. رقمهای والدینی ساحل، ورامین، گلستان و خرداد به ترتیب مناسبترین والدین برای بهبود کیفیت الیاف، وزن غوزه و تعداد غوزه بودند. میزان هتروزیس عملکرد هیبریدها ارتباطی با پتانسیل والدین نشان نداد، ولی با میزان اختلاف والدین و ترکیبپذیری خصوصی (SCA) همبستگی نسبتاً قوی داشت و در بین ژنوتیپهای والدینی، بیشترین هتروزیس مربوط به تلاقی ژنوتیپهای داخلی و خارجی بود.

ارزیابی خلوص ژنتیکی بذر پنبه رقم ورامین با استفاده از خصوصیات ریخت شناختی
درپی مشاهده اختلاط ژنتیکی تودههای بذر پنبه طبقات سوپرالیت و الیت رقم ورامین تولید شده در استان خراسان رضوی ناشی از دو نوع خارج از تیپ بهصورت بذرهای تقربیاً بدون کرک و با کرک سبز رنگ درحد بالای استاندارد ملی بذر درسال1393 و بهمنظور ارزیابی خلوص ژنتیکی بذر،آزمایشی مزرعهای در سالهای1394 و 1395 در مزرعه تحقیقاتی ستاد مؤسسه تحقیقات ثبت و گواهی بذر و نهال در کرج به اجرا در آمد. بدینمنظور در سال اول بذرهای خارج از تیپ تقربیاً فاقد کرک و با کرک سبز رنگ و تیپ کرکدار و با کرک سفید و توده بذر تولید شده که واجد بذرهای تیپ و خارج از تیپ رقم ورامین از طبقات سوپرالیت و الیت بود بهصورت دو آزمایش مزرعهای جداگانه برپایه طرح پایه بلوکهای کامل تصادفی با چهار تکرار کشت شدند. سپس، ارزیابی خصوصیات ریختشناختی(مورفولوژیک) بوته مطابق دستورالعمل ملی آزمونهای تمایز، یکنواختی و پایداری(DUS) پنبه تتراپلوئید انجام گرفت. هرگونه تغییرات این خصوصیات نسبت به تیپ ثبت شده خصوصیات ریختشناختی مذکور تعیین گردید و در مرحله شکفتگی کامل غوزهها تعداد بوتههای خارج از تیپ تقربیاً فاقد کرک و با کرک سبز رنگ هر کرت شمارش و تعیین شدند. همچنین وش کرتها برداشت شده و عملکرد وش و سپس خصوصیات تکنولوژیکی الیاف شامل: نسبت یکنواختی الیاف (UR)، طول الیاف بلندتر از50 درصد الیاف (UHML)، ظرافت الیاف(شاخص میکرونرMI)، استحکام و انعطافپذیری الیاف تعیین شدند. در سال دوم، آزمایش به همین صورت برای بذرهای نتاج که به ترتیب از طبقات الیت و گواهی شده بودند تکرار شد. نتایج نشان داد، بروز بوته و بذرهای خارج از تیپ مشاهده شده تحت تأثیر محیط قرار نداشت. همچنین درطی دو سال اجرای آزمایش جمعیت بوتهها و بذرهای خارج از تیپ افزایش یافت. با افزایش اختلاط در طی سالهای آزمایش کمیت و کیفیت محصول تولیدی کاهش یافت. بهطوریکه عملکرد وش و کیل بهعنوان مهمترین شاخصهای کمی تولید محصول و خصوصیات کیفی الیاف بررسی شده با افزایش شدت اختلاط کاهش قابل ملاحظهای یافتند.

ارزیابی برخی از واکنشهای فیزیولوژیک ارقام حساس و متحمل پنبه در شرایط تنش خشکی
ارزیابی واکنشهای فیزیولوژیک متفاوت ارقام پنبه در شرایط تنش خشکی، روشی دقیق و مطمئن جهت پایش روابط گیاه و آب بوده و میتواند در جهت اصلاح ارقام متحمل به خشکی بکار رود. لذا این پژوهش برای ارزیابی مقاومت روزنهای، دمای برگ، کانوپی و همچنین حداکثر کارایی فتوسیستم (ΔF/F́m) IIبر روی چهار رقم پنبه حساس (کوکر349 و نازیلی84) و متحمل (ارمغان و ورامین) به خشکی انجام گرفت. ارقام پنبه به عنوان کرتهای فرعی در سه سطح آب مصرفی به عنوان کرتهای اصلی (آبیاری به میزان (I33%) 33%، (I66%) 66%، و (I100%) 100% نیاز آبی) به صورت کرتهای خرد شده، در قالب بلوکهای کامل تصادفی در سه تکرار در مزرعه ایستگاه تحقیقات کشاورزی کاشمر در سال 1390 مورد ارزیابی قرار گرفتند. نتایج نشان داد به دنبال تنش خشکی، میزان افزایش مقاومت روزنهای ارقام متحمل بیشتر از ارقام حساس در شرایط بود. همچنین تنش خشکی سبب افزایش معنیدار دمای برگ (3/15 درصد) و کاهش معنیدار حداکثر کارایی فتوسیستم II (8/14 درصد) ارقام پنبه شد. علاوه بر این شیب تغییرات دمای برگ با افزایش مقاومت روزنهای در ارقام حساس بیشتر از ارقام متحمل بود. نتایج همچنین بیانگر تفاوت معنیدار ارقام از نظر حداکثر کارایی فتوسیستم IIبود. در این ارتباط رقم متحمل ورامین از بالاترین و رقم حساس کوکر 349 از کمترین حداکثر کارایی فتوسیستم IIبرخوردار بودند. بطور کلی در شرایط تنش خشکی واکنشهای فیزیولوژیک موثر بر رشد و عملکرد در ارقام متحمل به میزان کمتری در مقایسه با ارقام حساس تحت تاثیر قرار میدهند.

بررسی فنولوژی و عملکرد ارقام لطیف و گلستان در پنبه (Gossypium hirsutum L.) تحت تاثیر تاریخ کاشت و کود نیتروژن
پنبه بهعنوان یکی از محصولات زراعی مهم در کشور از جایگاه ویژهای برخوردار است. آزمایشی مزرعهای در سال 1395 در مزرعه تحقیقاتی مرکز تحقیقات پنبه کشور واقع در استان گلستان، شهر هاشمآباد به صورت اسپلیت پلات فاکتوریل در قالب طرح بلوکهای کامل تصادفی انجام شد. تیمارهای آزمایش شامل تاریخ کاشت (8 تیرماه و 21 تیرماه) بهعنوان فاکتور اصلی، و ترکیب فاکتوریل رقم (لطیف و گلستان) و کود نیتروژنی (شامل سه سطح : مطلوب، یک سوم حد مطلوب و بدون کود) بودند. سطوح کودی در حالت مطلوب حدود 160 کیلوگرم در هکتار، و در حالت یک سوم حدود 53 کیلوگرم در هکتار بود. نتایج نشان داد اثر تاریخ کاشت و سطوح مختلف کود نیتروژنی در اکثر صفات فنولوژیک و عملکرد ارقام لطیف و گلستان معنیدار (α= 0/05) بود. رقم گلستان در تاریخ کاشت اول (8 تیرماه) و سطوح کودی 160 کیلوگرم در هکتار از بیشترین عملکرد وش، عملکرد دانه، وزن خشک قوزه و تعداد قوزه در متر مربع (به ترتیب 6/3868، 1/2532، و 8/4265 کیلوگرم در هکتار و 12/128 قوزه در متر مربع) برخوردار بود. بیشترین عملکرد الیاف و تعداد قوزه در متر مربع نیز در سطح کودی شاهد (با عملکرد الیاف 7/782 کیلوگرم در هکتار و 3/72 قوزه در متر مربع) مشاهده شد که با افزایش سطح کودی از صفر به 160 کیلوگرم در هکتار کاهش یافت که دلیل این امر اثر کود نیتروژن بر افزایش رشد رویشی ارقام پنبه و تاخیر در رشد زایشی بود. نتایج نشان داد در تاریخ کاشت اول ارقام لطیف و گلستان به ترتیب 118 و 126 روز پس از کاشت وارد مرحله رسیدگی فیزیولوژیک (باز شدن قوزه) شدند در حالی که در تاریخ کشت دوم ارقام پنبه پس از 141 روز وارد مرحله رسیدگی فیزیولوژیک شدند که احتمالا یکی از دلایل عملکرد بالای ارقام در تاریخ کشت اول زودرسی ارقام بود.
